Title: DNA Methylation Patterns of LINE-1 Specific to HC21p in Prostate Cancer Cells

Project Description: Long Interspersed Nuclear Elements (LINEs) are retrotransposons. They function by making a cDNA copy, reverse-transcribing it into DNA and reinserting itself back into the genome; thereby, increasing its copy number. Sequencing of the human genome revealed that LINEs are underrepresented on the short arm of Human Chromosome 21 (HC21); however, the ones that are there, are active. The short arm of HC21, is primarily composed of heterochromatin and contains active L1s. Several studies investigate the role LINEs in cancer; specifically, their methylation patterns. These studies are typically done in euchromatic regions. Because of repetitive sequences, heterochromatin is extremely difficult to sequence. However these regions can be targeted for study since they pay an important role in chromosome function and a few genes may be concealed in them. Here we investigate DNA methylation patterns in promoter regions of six full length L1’s in the heterochromatic region of HC21. We also compare locus specific L1 methylation on the short arm (inactive) to the long (active) These are carried out in prostate cancer cells with various forms of aggression.
